Abstract:
The small size and transparency of many teleost brains provide the unique opportunity to image from the entire brain in an individual animal. The application of whole-brain imaging in zebrafish has led to the generation of computationally derived neuroanatomical atlases and in vivo functional imaging that have provided unprecedented insight into neural connectivity and function within the vertebrate brain. Recently, many of these approaches have been applied to the Mexican tetra, Astyanax mexicanus, to investigate how neural circuits evolve in closely related populations of the same species. Here, we describe protocols for immunohistochemistry and image registration and in vivo functional imaging in transgenic fish harboring pan-neuronal GCaMP. These approaches allow for the direct comparison of neuroanatomical and neurophysiological changes between individual populations of A. mexicanus that have experienced different selective pressure and harbor many behavioral and physiological differences.
